« » Aluminium effect on activity of some antioxidant enzymes in buckwheat leaves Smirnov Oleksandr Evgenovych Taras Shevchenko National University of Kyiv, Kyiv, Ukraine, plantaphys@gmail.com There is an evidence that aluminium toxicity can cause excessive generation and/or accumulation of reactive oxygen species (ROS). To date extensively accepted that ROS are responsible for biopolymers various stress-induced damage and disrupt majority cellular mechanisms in plants. Superoxide dismutase (SOD, EC 1.15.1.1) and catalase (CAT, EC 1.11.1.6) together with low-molecular mass antioxidants act as the main defense against ROS produced in various parts of plant cells. Phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) is a key enzyme in synthesis of phenolic compounds equipped in plants possess of antioxidant activity. Seeds of common buckwheat (Fagopyrum esculentum Moench.) were germinated in the dark at 25°C in Petri dishes with deionized water. Plants were grown in sand culture in controlled conditions. After 7 days the nutrient solution was contributed by aluminium (50 µM). Each day the solutions with Al were adjusted to a pH of 4.5. For investigation of enzyme (SOD, CAT and PAL) activity were used 17 day leaves of controlled and treatment plants. Both SOD and CAT are important enzymes associated with antioxidant defense system in plant cells. Compared to the control, the SOD activity was increased by 2 times in treatment leaves. The CAT activity was inhibited with no significant difference (p 0.05) ­ 4%. PAL showed a significantly higher activity under aluminium toxicity ­ by 2.5 times higher relative to control level. This investigation showed that the increases of SOD and PAL activity, not significant decrease of CAT activity could be an important part of common buckwheat resistance mechanisms to aluminium toxicity, such as regulation ROS balance, prevent the oxidative damage effects in plant cells.
